These volumes present the results of two studies undertaken for the Directorate General of Employment, Social Affairs and Education of the Commission of the European Communities.

The aim was to review available data concerning the metabolism of a selection of toxic chemi­ cals in man and to develop mathematical models representing this meta­ bolism.

Volume 1 is the report from the first study and presents re­ views of the data for each of the substances and proposes models for their metabolic behaviour.

Volume 2 is the report of the second study and contains a review of new data relevant to the development of meta­ bolic models for each substance.

The modelling of uptake and retention by the two major routes of uptake (the respiratory system and the gas­ trointestinal tract) is also reviewed.

The implementation of the models in a computer code is described and results are presented illus­ trating organ concentrations and rates of excretion for typical regimes of exposure. vi 1 Foreword This report contains the results of a study undertaken by Associated Nuclear Services (ANS) for the Directorate General of Employment, Social Affairs and Education of the Commission of the European Communities (CEC).

This study was entitled "Models for the Metabolism of Chemical Carcinogens in Man". vii 2 Summary This report includes results of a review of the metabolism of selected actual and potential human carcinogens and proposals for pharmacokinetic models to represent that metabolism.
